Comment: CC-406 is a good choice for a cell-wall deficient strain in mt-. An even better one, however, is CC-3491, which was derived from CC-406 by a series of four back-crosses to wild-type. Selection for a strain that mates well also ensures that most cells in the population are flagellated.
